Introduction

RAD 140, also known as Testolone, is an orally bioavailable selective androgen receptor modulator (SARM) that is currently being developed by the pharmaceutical company Radius.

The initial reason behind the drugs creation was to aid as a therapeutic means to treat breast cancer and muscle wasting.

As this SARM only began development in 2010, studies that analyze its physiological effects are very limited.

A lot of the current information on the drug is based on anecdotal user reviews, which are generally quite positive, especially regarding its ability to aid muscle growth.

There are also numerous reports of enhanced speed and stamina during workouts when supplementing with RAD140.

Does RAD140 Work?

RAD140 is Tissue-Selective

RAD140 has shown to be tissue-selective, where it stimulates the activity of androgen receptors without causing irregularities in the activity of other tissues such as the prostate and breast tissue [1].

More evidence is needed but this would mean that RAD140 does not convert to estrogen or DHT at any substantial rate.

At least on paper this makes it a drug with steroid-like effects but without as many serious side effects.

In fact, data shows that RAD140 does not just avoid estrogen receptors, but it appears to suppress their activity. This is actually one of the reasons why RAD140 may present a therapeutic approach for treating breast cancer [2].

May Reduce Brain Damage

Interestingly, the loss of androgens from normal aging processes can negatively impact androgen-responsive tissues, including the brain, and is significant risk factor for the development of neurodegenerative disorders including Alzheimer’s [3].

Androgens induce numerous beneficial and protective neural effects, including the promotion of synapse formation and neurogenesis, which contributes towards the survival of brain cells [4].

A great finding for RAD140 is that it can even provide considerable neuroprotection against neurotoxins that would normally induce minor brain damage [5].

This has also been found in lab studies, with RAD140 demonstrating its effectiveness at reducing brain cell death from internal damage. Mechanistically, this neuroprotection is mainly due to MAPK signaling. However, it is also hypothesized that RAD140 systematically activates glutamate receptors in the brain which play a key role in neuronal cell death (specifically in the hippocampus).

These novel findings demonstrate preclinical efficacy of RAD140 in neuroprotective actions against Alzheimer’s disease and related neurodegenerative diseases [6].

May Prevent Tumor Growth

Preclinical studies in multiple in animal models indicate that RAD140 has potent anti-tumor activity within certain cancers, such as the breast.

These animal studies were convincing enough for researchers and begin to study this within humans as an anticancer agent, with the first clinical tests being conducted at this time [7].

Not much else can be said on the link between RAD140 and tumor growth until research has been finished, but it is certainly interesting to note such an important potential benefit.

May Increase Muscle Mass

Based on hundreds of online reviews, using RAD140 will inevitably aid muscle growth and athletic performance.

However, interestingly, this has not yet been tested in scientific studies.

Due to the similarities between RAD140 and other SARMs, such as ostarine, it is very likely that RAD140 is able to improve an individuals body composition.

More research is needed to confirm this, and to analyze the degree to which it might benefit have such an effect.

RAD140 Safety And Side Effects

Current research on RAD140 has indicated that it is generally a safe drug when taken at those doses that have been studied (<5mg per day).

A key finding is that it does not appear to negatively impact the liver, which is one of the major concerns with steroid use [8].

However, as it is still in early developmental stages, this does not mean that there are no long-term side effects.

Based on subjective online user reviews, some side effects that could be experienced include headaches and insomnia.

Is RAD140 Legal?

RAD140 is not currently approved for human use or consumption in any country.

The drug can be purchased through non-pharmaceutical companies that create their own version of the drug, but safety and quality cannot be guaranteed.

World anti-doping organizations also clearly state that RAD140 is not allowed to be taken by athletes who are part of drug tested sports.

This is important as RAD140 has been found in many over-the-counter supplements due to contamination or a lack of manufacturing quality protocols. For example, in May 2017, Dynamic Technical Formulations had to recall one of their products as the FDA had found traces of this drug within the product.

Conclusion

RAD140 is a SARM that has been developed to treat breast cancer and muscle wasting.

There is a lack of human studies on this drug, but preclinical trials have shown it may be beneficial for inhibiting tumor growth and reducing brain degeneration from aging.

Online reviews clearly indicate that RAD140 has the ability to help build lean muscle mass, but this is yet to be tested in the research.

The safety of RAD140 is also not known at this point, and human trials are necessary to further analyze its efficacy to treat certain conditions.
